![]() It has always been, most specifically, an ‘all-road’ race. Only after a short hiatus, which coincided with the gravel boom in the US, did the race fall in line with the discipline of gravel, even if the course did not change much. In fact, it stood as a road race for a long time, even flirting with professional status and team cars at one point. Originally conceived in 1999, Rouge Roubaix is not a pure gravel race. Deep in the backwoods around Saint Francisville, a hidden gem of American racing thrives: Rouge Roubaix. Louisiana might not be the first place one thinks of when talk turns to gravel, but down amongst the bluffs and bayous of the Mississippi River is a collection of small gravel roads that offer a fascinating playground for any gravel enthusiast. The race was a gravel event back in 2015 when gravel looked a bit different Yes, those are 25mm rim brake tyres on a rim brake road bike. Every year the course changes slightly, but it all generally visits similar territory, so the ratio of 80 miles to 8,500 feet of climbing is a good reference point to work from. ![]() There are a couple sections of singletrack with a deceptively large amount of exposure which induce a level of thrill to the proceedings. That terrain provides constant challenges with inconsistent surfaces, grass, sand and mud all in play. ![]() The majority of the race runs on two-track or singletrack trails through the foothills of the southern Sierras around Bakersfield. Given that the area doesn’t usually get much tourism, the event has been wholeheartedly embraced by the rural community, which gives it a fantastic community feel.Īs for the racing? Expect a little of everything. Furthermore, since growing in popularity, the race has begun attracting riders from the big Southern Californian cities. That canvas makes for a delectable-looking course for riders from all over to take on. Yet for a couple of months out of the year, with the wetter weather of the winter offering respite from the dry summers, Bakersfield springs to life and becomes undeniably beautiful for cycling in the rolling green hills of the area. The city, which is one of California’s gruffest working-class towns, is for many just a waypoint on the way to Los Angeles, Las Vegas or Phoenix. Led by race director Sam Ames, the self-described “Chief excitement officer and ringmaster of the shit show”, the event is “less of a race, and always an adventure and gravel-ish party”.Ī fascinating aspect of the race, as is a common theme in gravel, is how the event ended up in its hometown of Bakersfield. Beyond the living room fly-by, riders should expect a fun-forward event that never takes itself too seriously. Yet, the race in Bakersfield, California is so much more than that one quirky element. At the Rockcobbler, someone actually does, with the race course running through a very gracious host's living room. Small gravel events are often akin to someone welcoming you into their home. ![]()

![]() The widespread use of new technologies and the search for innovative learning environments that connect with the current interests of students has led to the use of dynamic geometry programmes such as SketchUp. In this article, we focus on the evaluation the students carry out of the usefulness and degree of satisfaction of the use of the SketchUp software for their training in the space and geometry domain (SGD) through implementing the workshop. Our intention was to generate the need to coordinate logical-mathematical knowledge, as well as its learning and teaching, in a realistic context linked to the students' future professional activity. Said activity is organised in a workshop called We build our ideal school, in which we replaced the traditional use of manipulative material (Polydron, Geomag, Multicubes, etc.) by the use of the SketchUp design software. To illustrate this reflective process, one of the activities implemented in the subject called development of mathematical knowledge in Early Childhood Education, taught on-line in the third year of the Early Childhood Education (ECE) degree, will be presented. All this has entailed a deep reflection in search of new procedures to address, amongst many others, the following questions ( Esteve Ruescas and Alsina, 2020): how to plan active learning activities that encourage enquiry and promote the development of the didactic-mathematics competence of students in on-line learning environments? How to replace the use of manipulative materials? How to ensure student motivation and performance? During those activities, how to manage and mediate, encouraging communication, discussion and negotiation between students and with the teacher?Īlthough we have tried to keep the meaning and educational value of the subjects intact, the process of reflection has led to a significant reconstruction of the activities, strategies and methodological actions we had been using in previous courses. The ways of communication with and between students have changed completely and the classroom context makes the use of manipulative materials impossible. Training teachers in the use of ICTs is therefore key in the development of digital competences. The use of ICTs in themselves does not add value to the training process, it is the use the teacher makes of them that provides them with value. Those competences can help them when incorporating ICTs in their future teaching and professional practice ( Tokmak et al., 2013). Incorporating new technologies is also an opportunity for training in digital competences that future mathematics teachers should possess ( Stein et al., 2020). More importantly, it allows us to explore how these resources can be integrated to create an educational proposal that has a significant impact on the professional development of our students. ![]() ![]() However, this supervening situation offers us the opportunity to enquire about which ICT resources are potentially interesting for our training purposes. Higher Education has not been spared either, and has had to adapt fast to blended and on-line training contexts where the use of new technologies has been imposed as a non-negotiable need. The circumstances of the COVID-19 pandemic, which we are currently experiencing worldwide ( Hodges et al., 2020 Porlán, 2020 UNESCO, 2020), affect all areas of society, and generate important social, personal and economic difficulties. roofed with windows and doors fixed.Īll indicators show the census results are in line with expectations based on the benchmark values used to assess coverage of the 2021 PHC. ![]() The voltage is very important, as a high voltage can overheat and distort the bands. The gel is then connected to the power supply and allowed to run. ![]() The samples and a marker are loaded into the wells, and the empty wells are loaded with sample buffer. Gels are usually made by pouring them between two glass or plastic plates, using the solution described in the protocol section. The proteins when loaded on the gel have a negative charge, as they have been denatured by heating, and will travel toward the positive electrode when a voltage is applied. Protein is thus separated by their size more so in this gel, as the smaller proteins to travel more easily, and hence rapidly, than larger proteins. The lower gel, called the separating, or resolving gel, is basic (pH 8.8), and has a higher polyacrylamide content, making the gel's pores narrower. The higher, stacking gel is slightly acidic (pH 6.8) and has a lower acrylamide concentration making a porous gel, which separates protein poorly but allows them to form thin, sharply defined bands. Western blot uses two different types of agarose gel: stacking and separating gel. A negative control is a null cell line, such as β-actin, is used as well to confirm that the staining is not nonspecific. This helps to confirm the identity of the protein, and the activity of the antibody. For a positive control a known source of target protein, such as purified protein or a control lysate is used. It is also very important to have positive and negative controls for the sample. Denaturing the high structure ensures that the negative charge of amino acids is not neutralized, enabling the protein to move in an electric field (applied during electrotransfer). The sample is heated after being diluted into a loading buffer, in order to denature the higher order structure, while retaining sulfide bridges. A tracking dye (bromophenol blue) is also present in the buffer allowing the researcher to see how far the separation has progressed. Using this concentration allows to measure the mass of the protein that is being loaded into each well by the relationship between concentration, mass, and volume.Īfter determining the appropriate volume of the sample, it is diluted into a loading buffer, which contains glycerol so that the samples sink easily into the wells of the gel. Protein concentration is often measured using a spectrophotometer. This eventually allows the researcher to ensure that the samples are being compared on an equivalent basis. Since tissue sample display a higher degree of structure, mechanical invention, such as homogenization, or sonication is needed to extract the proteins.Īfter extracting the protein, it is very important to have a good idea of the extract's concentration. ![]() This should be done in a cold temperature with protease inhibitors to prevent denaturing of the proteins. Protein extraction attempts to collect all the proteins in the cell cytosol. This will be followed by the theoretical explanation of the procedure, and in the later section, troubleshooting tips for common problems.Ĭell lysates are the most common form of sample used for western blot. The paper will first describe the protocol for western blot, accompanied by pictures to help the reader and theory to rationalize the protocol. The thickness of the band corresponds to the amount of protein present thus doing a standard can indicate the amount of protein present. As the antibodies only bind to the protein of interest, only one band should be visible. The bound antibodies are then detected by developing the film. The unbound antibody is washed off leaving only the bound antibody to the protein of interest. The membrane is then incubated with labels antibodies specific to the protein of interest. These results are then transferred to a membrane producing a band for each protein. In this technique a mixture of proteins is separated based on molecular weight, and thus by type, through gel electrophoresis. Western blot is often used in research to separate and identify proteins. Therefore, less form-fitting around the hands can be loose-fitting and uncomfortable to wear and are not well-suited for precise tasks. On the contrary, vinyl gloves are less elastic. įor instance, latex gloves fit snugly around your hand like a second skin they are super comfortable to wear.įurthermore, they offer better dexterity and grip for sensitive and precise procedures. Likewise, elasticity directly contributes to comfort. In other words, they have low molecular cohesion, are less elastic, and are much easier to break and tear. Meanwhile, the mechanical strength of vinyl gloves compared to latex gloves is much lower.īecause vinyl molecules don’t stick well together ![]() They are considered the most elastic type of gloves. Latex Gloves… Which is more comfortable?Įlasticity is the ability of the gloves to return to their original shape after being pulled and stretched.Īs close as human skin, latex gloves have an extraordinary ability to stretch. To summarize, natural rubber latex gloves come from rubber trees, while synthetic rubber vinyl gloves are made from PVC polymer s.Įach goes through chemical and mechanical processes, from raw materials to disposable gloves we know to have the properties we will discuss next. Polyvinyl chloride is the world’s third-most widely produced synthetic polymer of plastic after polyethylene and polypropylene. ![]() These plasticizers make the PVC softer, pliable, and moldable enough to be transformed into gloves. However, it can be turned into a flexible material by adding plasticizers called phthalates. PVC is usually rigid and used in pipes, plastic bottles, etc. PVC is a petroleum-based polymer synthetically made by merging monomers of ethylene and chloride in a series of chemical processes. Therefore, vinyl gloves are most often referred to as PVC gloves. Vinyl gloves are made from a raw synthetic material called polyvinyl chloride (PVC). Now let’s see the synthetic rubber vinyl gloves and what they are made of. To illustrate, Latex is also called natural rubber latex as it comes from the rubber tree and is sometimes called caoutchouc.Īlthough people might use the terms alternatively, they are different rubber is an elastic material made either naturally from Latex or synthetically from petrochemical materials such as PVC and neoprene. When a plant is injured, it secretes Latex to seal the tear and prevent further damage, like human blood clots. It is a milky white complex emulsion of proteins, sugars, oils, alkaloids, tannins, gums, and starches that coagulates when air exposure. Plants typically produce Latex as a defense against insects and injuries. It’s the most common latex source, yet you can find Latex in almost 10% of flowering plants. This tree originally came from Brazil but is now mainly cultivated in Thailand, Malaysia, Indonesia, and other southeast Asian countries as well as Africa. It’s a natural material (a protein) that comes from the rubber tree sap (also called Hevea brasiliensis). Similarly, it has been a rockstar for many years. Latex is the first known material to be used in gloves. Let’s compare the material of vinyl and latex gloves Each disposable glove is made from a different material.ĭifferent materials contribute primarily to the properties of the gloves. Are you in the food industry or the medical field? Last week, Hawaii’s DOT started to post recent traffic counts on its website, chronicling the steep dive to show how the virus is taking a toll on everyday life. The COVID-19 pandemic also offers a glimpse of what’s possible when employment isn’t so dependent on transportation, with so many people working from home these days. This unprecedented pandemic shows just how well traffic conditions can illustrate a tanking economy. “Everything has just nosedived,” he added. Cities like Los Angeles that are notorious for traffic have no traffic. ![]() Nationwide, “we’ve never seen anything like what we’re experiencing right now,” said Trevor Reed, an analyst at the Kirkland, Washington-based traffic consulting firm INRIX.īy mid-March, the nation’s 25 largest metro areas - places usually snarled by traffic - were all in a “free-flow” during rush hour, Reed said. Meanwhile, on the neighbor islands, which rely more heavily on tourism, the traffic counts have decreased by more than half in some places. “You see it every year when UH is out of session,” Sniffen said. But transportation experts say a mere 5% reduction is the difference between gridlock and smoothly flowing traffic. Maybe you thought the number would be higher, given the circumstances. The state’s Department of Transportation reports that daily traffic counts are down nearly 50% along some of the H-1 freeway’s most heavily traveled corridors compared to last year. However, a lot of those missing cars represent the staggering 160,000-plus jobless claims that Hawaii has seen as of Thursday, just a few weeks into the pandemic crisis. Public health experts say that will help ease the strain on hospitals, giving those people severely stricken by the respiratory disease a better chance to recover. The clear roads suggest Oahu residents are largely following state and county orders to stay home in attempts to slow the spread of COVID-19. To be sure, that’s not entirely a bad thing. Instead, the traffic cruised along faster than I’d ever witnessed at that hour. Normally, that spells doom for west-siders trying to get into town. ![]() On a recent weekday morning, it took less than 20 minutes to get from Kapolei to the Vineyard Boulevard exit on the H-1. “It’s a horrific situation, where all of us will know somebody who gets affected by this because they lost their job or lost hours.” “This is definitely not the way we want to get to a free-flow condition,” said Ed Sniffen, deputy director for the state Highways Division. Turns out a global pandemic involving a highly contagious virus - one that has prompted mass quarantine and erased thousands of local jobs - was not a great way to solve Oahu’s traffic woes. Yet driving around the island these days is eerie and somber. It has never been easier to get into town in the morning and then get home to your family at night.
